Gianluca Buonanno was born in Borgosesia, Italy. At sixteen, Buonanno joined the Italian Social Movement. He graduated in accounting at the Technical Commercial Institute of Romagnano Sesia in 1987. He worked for a few years as a sales assistant.

In 1990, he was elected municipal councillor of Serravalle Sesia for the Italian Social Movement - National Right. He was elected mayor in 1993 in a civic list, which he was re-elected to in 1997. He was provincial councillor for Alleanza Nazionale (and then for the Lega Nord) in Vercelli from 1995 to 2009 and was vice-president of the province between 2007 and 2009. Having become ineligible for mayor of Serravalle Sesia after two serving consecutive terms, he ran as an independent (Lista Buonanno) in the Italian general elections of 2001 in the majority constituency of Cossato.

In 2002, he left Alleanza Nazionale and joined the Lega Nord. He was elected mayor of Varallo Sesia (Vercelli province) in 2002 and would be re-elected to this seat in 2007. In 2009, he left the post of vice-president of the Province of Vercelli and was appointed councillor for the municipality of Borgosesia.

In 2010, he was elected for the first time in the Regional Council in the Vercelli constituency. He was a deputy in the Chamber in the XVI Legislature of the Piedmont 2 constituency. In October 2011, he expressed to the Administration of the Chamber of Deputies the formal renunciation of a possible future parliamentary annuity. On 25 February 2013, he was re-elected to the Chamber of Deputies in the Piedmont 2 constituency and joined the Board of Directors of the Lega Nord parliamentary group.

Buonanno was elected a Member of the European Parliament for the North-West Italy Constituency on 25 May 2014. He was a non-affiliated member before joining the Europe of Nations and Freedom Group. He was a member of the Committee on Industry, Research and Energy and the Delegation to the ACP-EU Joint Parliamentary Assembly. He was a substitute member of the Committee on Agriculture and Rural Development and the Delegation to the EU-Russia Parliamentary Cooperation Committee.
